KATP channels in metabolic sensing: Mechanistic insights from CryoEM structures
KATP channels are ligand-gated potassium channels that couple cellular metabolism with membrane potential to regulate a broad range of cell activities: from hormone secretion, vasodilation, protection of cardiac and neuronal tissues against ischemic injuries, to learning and memory. Each KATP channel is composed of four inward rectifying potassium channels, Kir6.1 or Kir6.2, which form the ion-conducting pore, and four regulatory sulfonylurea receptors, SUR1, SUR2A, or SUR2B, from the ABC transporter family. Mutations in genes encoding KATP channel subunits are linked to several human diseases, including hyperi...

Revealing Structural Polymorphism in ATTR Amyloidosis: Insights from cryo- Electron Microscopy ATTR amyloidosis results from the self-assembly of amyloidogenic transthyretin (ATTR) into amyloid fibrils that deposit in tissues, leading to organ failure and death. This conversion is driven by mutations in ATTRv amyloidosis or aging in ATTRwt amyloidosis. While ATTRv amyloidosis exhibits extreme p...

Cryo-EM structure of HIV-1 Rev Response Element RNA: Structural basis of Rev-RRE interaction Viral genomic RNA not only codes for proteins but also acts as a regulatory element during virus replication and pathogenesis. While the primary RNA sequence determines viral protein structure and function, the secondary and tertiary structure of RNA are critical to its role as a regulatory element. Dur...

Using cryo-EM to understand dynamic interactions As a trained NMR spectroscopist and experienced X-ray crystallographer, I will provide our newly gained experience and insights from using cryo-EM as a routine tool in our laboratory. As an example, I will use a recent complex that we determined by cryo-EM: PP2A:B55-ARPP19. PP2A:B55-ARPP19 is critical as progression through the cell cycle is cont...

Engineered Protein Cages for Imaging of Small Proteins by Cryo-EM Recent technical advances have made cryo-electron microscopy (cryo-EM) an attractive method for atomic structure determination, but problems of low signal-to-noise prevent routine structure determination of proteins smaller than about 50 kDa. We have developed symmetric protein imaging scaffolds to display and solve the structure...

Have you ever looked at images from your microscope session and had no idea what you were looking at? Have you stared at fuzzy grey images until your eyes hurt? Interpreting micrographs, especially for non-ideal samples, is difficult for the un-trained eye. A panel of staff from the National Center for CryoEM Access & Training (NCCAT), the Pacific Northwest Cryo-EM Center (PNCC), and the Stanfo...
